Paxil® and Zoloft® - Birth Defects Linked to SSRI Antidepressants

Paxil® (paroxetine), Zoloft® (sertraline), and other popular antidepressants known as selective serotonin reuptake inhibitors (SSRI) have been linked to the serious birth defect Persistent Pulmonary Hypertension of the Newborn (PPHN), as well as other birth defects affecting the heart, lungs, brain, and nervous system, sometimes resulting in mental and physical disabilities.  In February 2006, The New England Journal of Medicine published a study showing that, under certain circumstances, pregnant women taking SSRI antidepressants could be more than six times as likely to give birth to an infant suffering PPHN.  PPHN is a condition where the blood of a newborn does not properly flow through the lungs, causing a lack of oxygen in the blood stream, often resulting in developmental delays and even death.  For more information, please see our Persistent Pulmonary Hypertension of the Newborn Information page.

SSRI antidepressant usage in the first trimester of pregnancy is also linked to birth defects.  In June 2007, The New England Journal of Medicine published another study, examining the use of different SSRIs, including Paxil® and Zoloft®, only in the first trimester.  This study concluded that individual SSRIs may increase the risks for specific birth defects.  For example, a pregnant woman who took Paxil® during the first trimester is 3.3 times as likely to have obstruction in the right ventricular outflow tract (a heart defect) and a pregnant woman who took Zoloft® during the first trimester is two times as likely to be born with a septal defect (a heart defect).  An analysis of the 2005 Swedish National Registry also showed that Paxil® usage in pregnancy can cause twice as many heart related birth defects.

Other birth defects that have been associated with SSRI usage, such as Paxil® and Zoloft®, incude:
  • Neural Tube Defects
  • Craniosynostosis
  • Omphalocele
  • Limb Reduction
  • Anal Atresia
  • Conotruncal Defects
  • Club Foot
  • Cleft Lip

Women who are pregnant or may become pregnant should consult with a physician if they are currently taking, considering taking, or considering discontinuation of Paxil® (paroxetine), Zoloft® (sertraline) or any Selective Serotonin Reuptake Inhibitor (SSRI) antidepressant.  Other common SSRI drugs include:
  • Prozac® (fluoxetine)
  • Lexapro® (escitalopram)
  • Celexa® (citalopram)
  • Effexor® (venlafaxine)
  • Symbyax® (olanzapine/fluoxetine)
  • Luvox® (fluvoxamine)
